This book brings together drug design practitioners, all leaders in their field, who are actively advancing the field of quantitative methods to guide drug discovery, from structure-based design to empirical statistical models - from rule-based approaches to toxicology to the fields of bioinformatics and systems biology. The aim of the book is to show how various facets of the drug discovery process can be addressed in a quantitative fashion (ie: numerical analysis to enable robust predictions to be made). Each chapter includes a brief review of the topic showing the historical development of quantitative approaches, a survey/summary of the current state-of-the-art, a selection of well chosen examples with some worked through and an appreciation of what problems remain to be overcome as well as an indication of how the field may develop. After an overview of quantitative approaches to drug design the book describes the development of concepts of "drug-like properties", of quantitative structure-activity relationships and molecular modelling, and in particular, structure-based design approaches to guide lead optimisation. How to manage and describe chemical structures, underpins all quantitative approaches to drug design and these are described in the following chapters. The next chapter covers the value of a quantitative approach, and also the challenge which is to describe the confidence in any prediction, and methods to assess predictive model quality. The later chapters describe the application of quantitative approaches to describing and optimising potency, selectivity, drug metabolism and pharmacokinetic properties and toxicology, and the design of chemical libraries to feed the screening approaches to lead generation that underpin modern drug discovery. Finally the book describes the impact of bioinformatics, current status of predicting ligand affinity direct from the protein structure, and the application of quantitative approaches to predicting environmental risk. The book provides a summary of the current state-of-the-art in quantitative approaches to drug design, and future opportunities, but it also provides inspiration to drug design practitioners to apply careful design, to make best use of the quantitative methods that are available, while continuing to improve them. Drug discovery still relies heavily on random screening and empirical screening cascades to identify leads and drugs and the process has many failures to deliver only a small handful of drugs. With the rapidly escalating costs of drug discovery and development together with spiralling delivery, quantitative approaches hold the promise of shifting the balance of success, to enable drug discovery to maintain its economic viability.
Comprehensive, yet practical and concise, the Oxford Specialist Handbook of Pacemakers and ICDs is the ideal training guide on how to implant, follow-up, and troubleshoot pacemakers and ICDs. Fully updated to include new technologies such as subcutaneous ICDs and MRI compatible devices, this new edition provides the latest guidelines and management strategies for the cardiology trainee and cardiac technician. Covering the principles, programming, potential complications, and troubleshooting for pacemakers, ICDs, and cardiac resynchronisation therapy, this title is an invaluable aid for anyone charged with providing or contributing to a pacing, ICD, or implantable loop recorder service. Written in a succinct bullet-point style, the second edition of the Oxford Specialist Handbook of Pacemakers and ICDs delivers key information in an accessible manner, with over 120 figures including x-rays and annotated ECGs to demonstrate pacing techniques and troubleshooting solutions.
Drug discovery is a constantly developing and expanding area of research. Developed to provide a comprehensive guide, the Handbook of Medicinal Chemistry covers the past, present and future of the entire drug development process. Highlighting the recent successes and failures in drug discovery, the book helps readers to understand the factors governing modern drug discovery from the initial concept through to a marketed medicine. With chapters covering a wide range of topics from drug discovery processes and optimization, development of synthetic routes, pharmaceutical properties and computational biology, the handbook aims to enable medicinal chemists to apply their academic understanding to every aspect of drug discovery. Each chapter includes expert advice to not only provide a rigorous understanding of the principles being discussed, but to provide useful hints and tips gained from within the pharmaceutical industry. This expertise, combined with project case studies, highlighting and discussing all areas of successful projects, make this an essential handbook for all those involved in pharmaceutical development.
Released in 2007 and the first handbook-sized practical training guide to provide easily accessible, detailed information on how to get good images, make key measurements and report findings, the first edition of Echocardiography rapidly become a bestselling echocardiography manual. Now in its second edition, and reflecting the most recent international cardiology and echocariography society guidelines, the handbook has been extensively updated throughout and features new sections covering 3D echocardiography, intracardiac echocardiography, contrast echocardiography, speckle tracking echocardiography and emergency echocardiography. In full colour throughout, the text is illustrated with over 300 images and comes with a DVD resource of over 175 image loops. Also including reference ranges to help with reporting and accreditation exam preparation, Echocardiography remains the essential reference for trainees undergoing accreditation and a daily source of information for those who perform clinical echocardiography.
A range of cardiac imaging techniques are available, each with a
unique approach. Most existing imaging books are predominantly
modality focused; however today's clinical cardiologist needs to
learn how to apply and integrate information from the different
modalities to aid clinical decision-making.
