When Evelyn, a beautiful Navy nurse, meets dashing ace Army fighter pilot Rafe, the two fall madly in love, only to be separated abruptly when he is called upon to help fight the war in Europe. Once in the air the plane is hijacked by convict Cyrus 'The Virus' Grissom, who gets rid of the crew of prison guards at the first scheduled stop, picking up more convicts as he does so. But US Marshal Vince Larkin is already on his trail. Poe does his best to stop Grissom from escaping with the group of convicts, and just to complicate matters further his diabetic cellmate reveals that he will die if he does not receive his next insulin injection within two hours.

There he meets the leader of the organisation, Dr. Silas Lamb (Kingsley), and one of his patients, Eliza Graves (Beckinsale), who Newgate becomes enamoured with. Shocked by the unconventional methods Lamb uses to treat his patients, Newgate decides to leave the asylum, determined to take Eliza with him.
