This book presents an overview of the most important current developments in the pre-analytical handling of tissue. It addresses in particular potential ways to improve the situation whereby methods employed in the pre-analytical phase – the period from surgical removal of tissue to the start of pathological processing – have remained essentially unchanged for decades with only modest standardization. It is examined how the pre-analytical period can be optimized, resulting not only in an increase in diagnostic quality but also in a reduction in processing time and costs. Among the key topics examined are the so-called cold ischemia time between tissue removal and fixation, the potential superiority of vacuum-based preservation over immediate formalin fixation, two-temperature fixation, molecular analysis methods, and the pre-analytics of specimens from particular tissues. The principal role of the diagnostic surgical pathologist is to serve a patient by assisting the clinician in charge of the patient's care. In that capacity,the pathologist provides the vital informationthat should be directly and indirectly useful in guiding the clinician toward the most appropriate therapy. The material pathologists receive most commonly is a biopsy or a part of an organ removed after a definitive pro- dure. To extract useful information, pathological evaluation proceeds according to a set of guidelines. Simply reporting a diagnosis of cancer is inadequate. Detailed additional information is needed so that the clinician can go on to establish a the- peutic plan as needed. To best serve the patient, close interaction between the cli- cian and the pathologist is vital. In the field of urological pathology, there are problems specific to this system. Typically, in the prostate, because of its location, there is little room for a surgeon to work to obtain adequate resection margins, unlike with many other organs. As a result, questions arise such as "What constitutes an adequate surgical margin?" "What is the significance of extraprostatic extension of neoplasm that is still inside the resection margin?" and "What is the significance of neoplastic glands found on the resection margin marked with the ink?" It has been shown that a prostate needle core biopsy can generate much information that is immediately useful in predicting the extent of cancer in the prostate and, consequently, the outcome for the patient.
Breast cancer is the most common cancer in females that accounts for highest cancer specific deaths worldwide. In the last few decades research has proven that breast cancer can be treated if diagnosed at early stages and proper therapeutic strategy is adopted. In this second volume in the series exploring Tumor Dormancy, Quiescence, and Cellular Senescence, discussion is focused on the role of tumor dormancy in diseases such as breast cancer, melanoma, prostate cancer, liver cancer and lung cancer. M. A. Hayat, the series editor, writes in the preface that little is known of factors regulating the transition of residual cancer into a dormant state or the subsequent reinitiation of growth. A majority of us, he says, have in situ tumors that may remain dormant or may progress into a lethal form of cancer; the former are prevented from recruiting their own blood supply. Section I covers Molecular Mechanisms, with chapters on the role of NAE inhibitor MLN4924; oncogene-induced senescence; the role played by mitogen-activated protein kinase in the induction of cellular senescence; mechanisms of premature cell senescence and other topics. Section II examines Tumor and Cancer, discussing defects in chromatin structure and diseases; the role of fibrosis in tumor progression and the dormant to proliferative switch; the function of ING proteins in cancer and senescence and more. The final section is devoted to Stem Cells and Cancer Stem Cells, featuring chapters showing that senescent-derived pluripotent stem cells are able to redifferentiate into fully rejuvenated cells; that the transcription factor Gata2 regulates quiescence in haematopoietic stem and progenitor cells; and discussing dormancy and recurrence of cancer stem cells in bone. The contributors point out that the quiescent state regulates hematopoietic stem cells and muscle stem cells, and detail the role of kinase in the mediation of reversible quiescent state in a subset of ovarian, pancreatic, and colon cancers. Molecular mechanisms underlying stress-induced cellular senescence and accumulation of reactive oxygen species and induction of premature senescence are also presented. Discussion includes the important role of microRNAs in oxidative stress-induced apoptosis and senescence and the effect of microRNA as a modulator of cell proliferation in lung cancer. The book includes an explanation of the suppression of cellular senescence in glioblastoma brain tumor. Taking a broad and varied perspective, this volume was written by 70 contributors representing 11 countries.

The incidence of cutaneous melanoma has risen considerably over the last few decades, and much attention is now paid to the early diagnosis of this tumour. Biopsy Pathology of the Bronchi is intended to provide the practicing pathologist with a convenient source of diagnostic information, combined with pertinent clinical patterns and a background of histogenesis. It is hoped that the book will facilitate the accuracy of diagnosis, minimize pitfalls of interpretation and assist in the rapid evaluation of acute and chronic bronchial lesions, including neoplasms. Biopsy is taken in the widest sense and the book includes discussions on cytology, microbiopsies (fine needle aspiration biopsies, curette samples, and biopsies obtained by microforceps) and macrobiopsies, obtained at the time of surgery. Part I of the book deals with the procurement and preparation of bronchial specimens and includes a chapter on fetal development and structure of the normal adult epithelium. In Part II, the histopathology of numerous bronchial diseases are described in detail. Emphasis is placed on the diagnostic features of human lesions' which are illustrated by numerous high quality light and electron micrographs.
