Sowohl die contractilen Eiweisse, wie die Mechanik und die Elektro- physiologie sind beim glatten Muskel weniger intensiv bearbeitet worden als beim Skeletmuskel. Deshalb sind gewisse Gebiete der Glattmuskelphysiologie gegenwartig noch mehr im Zustand einer qualitativen als einer quantitativen Beschreibung. Die Grunde fur die Vernachlassigung des glatten Muskels waren sicher lange Zeit in technischen Schwierigkeiten zu suchen. So ist es z. B. nicht leicht, fur jede Art glatten Muskels genugende Mengen zur chemischen Untersuchung frei von anderen Bestandteilen zu erhalten. Die Anwendung der Mikroelektrodentechnik, welche die Physiologie des quer- gestreiften und des Herzmuskels entscheidend gefoerdert hat, ist beim glatten Muskel schwierig, die Isolierung einzelner Fasern unmoeglich. Viel mehr noch als die methodischen haben wohl die gedanklichen Schwierigkeiten, welche bei der Betrachtung der einfachsten funktionellen AEusserungen des glatten Muskels auftauchen, die Untersucher abgeschreckt. Das kaprizioese Verhalten des glatten Muskels ist sprichwoertlich. Jeder Adept, der zum ersten Mal einen glatten Muskel seine Kontraktionen registrieren lasst, glaubt nicht, dass das Spiel des Schreibhebels etwas anderes als das Produkt wusten Zufalls sein koenne.

7 Diese Art der Informationsubertra- gung bietet eine Menge von Vorteilen, von denen hier nur drei erwahnt seien: 1. Verwendung eines einheitlichen biosynthetischen Mechanismus und nur weniger Bausteine (Aminosauren) zur Herstellung einer unubersehbaren An- zahl moeglicher Wirkstoffe. 2. Steuerung des Eintrittes der Wir- kung durch Ausloesung des (sehr schnell arbeitenden) Synthesemechanismus auf der Stufe der Gene oder aber durch proteolytische Freisetzung des Wirk- stoffes aus einer unwirksamen Speicher- form (z. B. Angiotensinogen, Abb. 8). 3. Aufhebung der Wirkung in kur- zester Zeit mittels gewoehnlicher proteo- lytischer Enzyme. Fur die therapeutische Anwendung von Polypeptidwirkstoffen bedingt Punkt 3 parenterale Verabreichung, weil die Stoffe im Magen-Darm-Trakt proteolytisch zerstoert bzw. schlecht resorbiert werden. Diesem Nachteil steht aber der nicht zu uberschatzende Vorteil gegenuber, dass beim Abbau des Wirkstoffes ausschliesslich physiologische Verbindungen (Aminosauren) entstehen. Somit ist jede Schadigung durch toxische Abbauprodukte, wie dies bei andern Pharmazeutika manchmal der Fall ist, ausgeschlossen. Im folgenden moechte ich als Beispiel hauptsachlich Wirkstoffe besprechen, an deren synthetischer Entwicklung meine Mitarbeiter und ich besonders beteiligt waren. Es sind dies das Gewebshormon Angiotensin und die Hypophysenhor- mone oc-Melanotropin, ss-Melanotropin und Corticotropin. In recent years the more physiological aspects of the cerebrospinal fluid have aroused interest, with the result that some progress in elucidating the mechanism of its formation and circulation, and its relationships with the central nervous parenchyma, has been made. The position is still very obscure, however, and it is the purpose of this review to collect the facts from a very scattered literature and to emphasize the gaps, in the hope that they will invite further research. In general, I have used as my starting point in time the years 1955 to 1956, the period when an earlier and more comprehensive review (DAVSON 1956) was going through the press; this has not preeluded the quota- tion of earlier work, however, when this was relevant. In writing this, I have of course been able to benefit by the series of articles in the Ciba Symposium held in London in 1957 and the scholarly reviews of the blood-brain barrier by BAKAY (1956) and DOBBING (1961). I have tried to be comprehensive, but the wide area of the literature over which the published work is scattered means, unquestionably, that I have overlooked many significant contri- 1 butions - 11.

53 catalytic interaction between enzyme-coenzyme complex and substrate (VENNESLAND and WESTHEIMER 1954; VENNESLAND 1955,1956; GUTFREUND 1959) have all been the subject of numerous excellent and exhaustive reviews. These topics, therefore, will not be treated in extenso here. The present summary will deal instead with more recent observations on the pyridine nucleotide dependent dehydrogenases which have necessitated a re-examination of their function and structure in the light of new information concerning their composition. The new compositional characterization of these enzymes dates from 1955 [VALLEE and HOCH 1955 (a), (b), (c)] when the DPN dependent alcohol dehydrogenase of bakers' yeast was identified as a zinc metalloenzyme. Since then several other DPN -dependent enzymes have been similady shown to contain zinc [VALLEE 1955, 1960 (a), (b)], and a number of DPN and TPN dependent dehydrogenases have been tentatively identified as metalloenzymes (VALLEE et al. 1956). Knowledge of the presence of zinc in these enzymes has provided a new set of parameters for the study of these systems. The metal in such systems retains much of the chemical reactivity exhibited in the ionic form, modified by its incorporation into the protein matrix. Since the metal is not only a compositional constituent, but is also involved in the protein structure and in the events of catalysis, its reactivity can be used to advantage in exploring specific enzymatic sites, and the general problems of biological catalysis.
