The aim of this book is to provide detailed protocols for studying
the molecular biology of the pathogen Mycobacterium tuberculosis,
and its int- actions with host cells. As established mycobacterial
laboratories move - wards exploiting the genome, and laboratories
with expertise in other fields apply them to mycobacteria, both
traditional and novel methodologies need to be reviewed. Thus the
chapters in Mycobacterium tuberculosis Protocols range from
perspectives on storage of strains and safety issues to the
application of the latest functional genomics technologies. The
last few years have been remarkable ones for research into M.
tuber- losis. The most important landmark by far has been the
completion of the genome sequence of the widely studied H37Rv
strain (1). We can now predict every protein and RNA molecule made
by the pathogen. This information is or will soon be enriched by
the addition of genome sequences of other strains from the M.
tuberculosis complex: a second strain of M. tuberculosis, My-
bacterium bovis, and the vaccine strain, M. bovis BCG. Valuable
comparative data will also be provided by the genome sequences of
Mycobacterium leprae, Mycobacterium avium, and Streptomyces
coelicolor. Another recent milestone for M. tuberculosis has been
the development of efficient mutagenesis me- odologies, the lack of
which has been a major handicap in functional studies.
"Re-energizing Citizenship" examines the dual character of civil
society. The malaise in civil society is seen as a key to
understanding anti-social behaviour, the weaknesses of regeneration
schemes and the divisions and antagonisms of our societies. Yet the
resources and skills hidden away in civil society, if they could be
unlocked, are seen as the key to effective intervention. The book
provides a critical examination of attempts to re-energize
citizenship in a range of contexts and offers insights into what
works.
From 1979 to 1997 Britain was a laboratory for experiments in local governance as the control and delivery of local services was switched from elected councils to appointed boards (guangos), private companies or self-management. This book is about four models of local governance: the traditional "localist" model, the New Right's "individualist" model, the New Left's "mobilization" model, and government's own "centralist" model. It tests them against public opinion as expressed in 2203 interviews with ordinary citizens, 788 with councilors, and 903 with members of appointed boards.
Confusion about governance abounds. Many lack appreciation of how
different traditions of thought in the social sciences contribute

issues head on, using a cross-disciplinary approach from political
science, public administration, international relations,
development studies, sociology and economics. Looking at collective
decision making in societal systems or organisations, the book
provides guidance on how power should be distributed, who should be
involved in a decision, and how policy should be enforced.
The growing interest in governance is precisely because established
institutional forms of governance appear under challenge and new
forms appear to be emerging. This reality is compelling new ways of
thinking about collective decision-making at a range of levels.
This book explores three critical areas of practice: environmental,
corporate and participatory governance. Knowledge of these
extensive intellectual resources will enrich academic debate and
enable those interested in reforming governance to move towards a
wider vision.
