This book deals with basic and clinical aspects of monoamine oxidase (MAO) subtypes A and B highlighting its importance in neurological and psychiatric diseases. Consequently the therapeutic actions of MAO-A and -B inhibitors in Parkinson's disease (PK) and depression are the focus of several chapters. As MAO is the basis of the development of the "oxidative stress hypothesis" of PD, several chapters are devoted to iron and iron-induced oxidative stress in various experimental studies and clinical conditions. Based on these findings, new compounds have been developed which not only block MAO, but are in addition, either inhibitors of acetylcholine esterase or have iron chelating properties. The aspect of "preclinical" and "clinical" neuro protection as well as MAO neuroprotection are additional topics covered in this book. MAO, iron and neuroprotection are seen in the framework of general anti Parkinson's therapy with chapters on levodopa, dopaminergic receptor agonists and clinical issues.

The aim of Early Markers in Parkinson's and Alzheimer's Diseases is to provide the reader with updated data on various approaches whose investigation and development could contribute to the discovery of early diag- nostic markers of these two degenerative diseases. Concerning Parkinson's disease, some of the topics dealt with in the book will help update the information previously reported in Early Diagnosis and Preventive Therapy in Parkinson's Disease. Concerning Alzheimer's disease, the scope and limitations of electrophysiological and brain imaging techniques with regards to early detection of the disease are documented. Various biochemical parameters, such as brain energy metabolism, levels of choline, and platelet monoamine oxidase activity are envisaged as some of the starting points for the discovery of early diagnostic markers of Alzheimer's disease.

Monoamine oxidase (MAO) is linked to psychiatric and neurological disorders, because inhibitors of the enzyme are used clinically for treatment of affective disorders and Parkinson s disease. One of the interesting new aspects of MAO is the occurrence in the human population of deletions of genes coding for one or the other enzyme subtype (A or B). This leads to the possibility of a genetic basis for psychiatric disorders based on MAO. Subjects with deletions of type A or B, and combined deletions, have been described. In the first group of 6 papers in this book, the occurrence and characterization of these phenotypes, as well as the structure of MAO genes, is explored. Advances in the biochemistry of MAO subtypes and their physiological function and localization in brain and periphery is included. Other sections of the book deal with the neuroprotective action of MAO inhibitors and their pharmacology, especially the pharmacology of new MAO-B inhibitors."

The role of the metals copper, zinc, magnesium, lead, manganese, mercury, lithium and aluminium in neuropsychiatric disease are well known and has been discussed on several occasions. Yet little attention has been paid to iron, the most abundant transitional metal in the body and the earth's crust. Iron plays a major role as a cofactor of numerous metabolic enzymes, it is important for DNA and protein synthesis, and has a crucial role in the oxygen carrying capacity of haemoglobin. Some of the most devastating diseases of systemic organs are associated with abnormal iron metabolism. Yet only very recently its role in the central nervous system has been considered. Thus nutritional iron defi ciency and iron overload afflict some 500-600 million people. It is also well recognized that too little or too much iron can produce profound effects on the metabolic state of the cell, and therefore the regulation of iron uptake and disposition is tightly relegated by the cell. Its transport into the cell and storage are handled by transferrin, ferritin and haemo siderin. Nowhere are these processes so well recognized as in the case of brain iron metabolism. Iron does not have ready access to the adult brain as it does to other tissues, since it does not cross the blood brain barrier (BBB). All the iron present in brain is deposited before the closure of BBB at an early age where it is sequestered and conserved. Therefore its turnover is extremely slow."

The Amine Oxidase Workshop was the brain child of Brian Callingham and Keith Tipton. The present book is the proceedings of the fourth in the series and in the past we have dedicated our meeting to individuals who have made significant fundamental contributions to the biochemistry, physiology and pharmacology of this interesting group of enzymes. The present meeting is not an exception because we have gathered here in Wtirzburg to honour an individual whose outstanding work on the autonomic pharmacology has span ned more than 30 years and whose contribution has made a vast impact in our understanding of the kinetic relationship between amine metabolism (that uptake and enzymatic inactivation) and adrenergic neurotransmission. We speak of course of Professor Ullrich Trendelenburg, or Ulli as he likes to be known. Like the amine oxidases, he has a unique place in Pharmacology. Professor Trendelenburg was born on the eve of the new year of 1922 in the city of Rostock in northern Germany, where his father was Professor of Pharmacology at the Faculty of Medicine of Rostock. His father accepted the offer to take over the corresponding chair in Freiburg im Breisgrau (in Southern Germany). As the youngest of four children, Ulli grew up mainly in the garden of the department, in the animal quarters and in the workshop. Of the various coworkers of his father, one he liked especially was Otto Krayer, the young pharmacologist."

Dementia of Alzheimer type (DAT), multiinfarct dementia (MID) and dementia occurring in the course of Parkinson's disease (PD +D) now make up one of the largest categories of chronic diseases in the elderly. In addition to the burden those illnesses impose on the affected individuals and their families they consume large socio-economic re- sources. In the light of all the above mentioned features, it seemed to us that a Symposium on behalf of the 125th Anniversary of Birth of Aloys Alzheimer was particularly well-suited to help to advance research on Alzheimer's disease and other dementias. This International Symposium combined with a Satellite Symposium about "Clinical Aspects of Alz- heimer Dementias" took place in June 1989 in Wiirzburg and has been organized by the Psychiatric Departments of the Universities of Wiirzburg and Munich (H. Beckmann, K. Maurer, P. Riederer, H. Hippius and H. Lauter) and the Department of Pathochemistry and General Neurochemistry of the University of Heidelberg (S. Hoyer). In the chapters which follow, thorough reviews of recognized authorities in the field of dementia are given in the four main fields of epidemiology, neuropathology, neurochemistry and clinics. This Symposium and the edition of this book would not have been possible without the generous support of E. Merck, Darmstadt. At the time when "Parkinson's Disease" is diagnosed in a patient, roughly two thirds of dopaminergic neurons of substantia nigra are already degenerated. The onset of the disease must, therefore, be much earlier. This book deals with early diagnosis and early preventive treatment which may sustain the process underlying the disease. By use of psychometric, kinesiologic, physiologic, histologic, biochemical, endocrinologic, pharmacologic and imaging techniques, including positron-emission tomography and brain mapping, specialists tried to focus on new diagnostic criteria. New methods including psychometric evaluation, apparative measurement of movement, analysis of peripheral blood and urinary constituents have supplemented this approach. It has been agreed that early preventive therapy consists of low dosis of L-DOPA plus benserazide, L-deprenyl and dopaminergic agonists.
