This book contains the refereed contributions from the 43rd annual meeting of ISOTT. The annual meetings of ISOTT bring together scientists from various fields (medicine, physiology, mathematics, biology, chemistry, physics, engineering, etc.) in a unique international forum. ISOTT conferences are a place where an atmosphere of interaction is created, where many questions are asked after each presentation and lively discussions occur at a high scientific level. This vivid interaction is the main motivation for members to participate and gain new ideas and knowledge in the broad field of oxygen transport to tissue. The proceedings include sessions covered various research topics including Multi-Modal Imaging/Spectroscopy & Instrumentation; Cancer Metabolism; Cellular Hypoxia and Mitochondrial Function; Brain Oxygenation and Function; Other Organ Function and Metabolism; Oxygen Transport in Sports, Diseases and Clinical Care; Acupuncture, Meridians, and Primo Vascular System; EPR, MRS and MRI.
Rabies is one of the oldest known pathogens, is incurable, and has the highest fatality rate of all infectious diseases. The Americas is the only region with bat rabies virus, including vampire bat rabies. The region is rich in cultural references and notable for many discoveries in the field, including the current vaccine potency test, diagnostic assay, conception of oral vaccines for wildlife, the first human survivor and the first successful canine rabies program executed at a broad level. Rabies remains the most important viral zoonosis, with tens of thousands of human fatalities and tens of millions of exposures annually, which can be used to model for other pathogens, such as COVID-19. There is an international effort to eliminate human rabies caused by dogs over the next decade, and the Americas represent the primary region with the greatest proof-of-concept evidence to accomplish this goal. This two-volume set addresses the medical history and modern results of rabies in countries throughout the Americas, including the implications of and on cultural, economic, sociological, and research developments in the region. Volume I presents an overview of concepts critical to the study of rabies in the region, including evolutionary aspects, reservoir ecology and control, elimination efforts, vaccine development, and disease hallmarks and progression. It also analyzes the long-term cultural, social, and economic impacts of the disease in the Americas.Â
Malaria is still a major global health problem, killing more than 1 million people every year. Almost all of these deaths are caused by Plasmodium falciparum, one of the four species of malaria parasites infecting humans. This high burden of mortality falls heavily on Sub-Saharan Africa, where over 90% of these deaths are thought to occur, and 5% of children die before the age of 5 years. The death toll from malaria is still growing, with malaria-specific mortality in young African children estimated to have doubled during the last twenty years. This increase has been associated with drug resistance of the parasite, spread of insecticide resistant mosquitoes, poverty, social and political upheaval, and lack of effective vaccines. This collection of reviews addresses many of these important issues of malarial immunity and immunopathology. They are of interest not only to malariologists, but hopefully also to the broader immunological community. Strong interactions with, and feedback from immunologists working in other infectious diseases and in basic immunology will help us to move the field of malaria immunology and therapeutic intervention forward more quickly.
This work in the field of tropical medicine provides an in-depth review in the development of vaccines against some of the most debilitating tropical diseases. This volume provides a state-of-the-art update on Fc Receptors (FcRs). It is divided into five parts. Part I, Old and New FcRs, deals with the long-sought-after FcR and the recently discovered FCRL family and TRIM21. Part II, FcR Signaling, presents a computational model of Fc RI signaling, novel calcium channels, and the lipid phosphatase SHIP1. Part III, FcR Biology, addresses major physiological functions of FcRs, their glycosylation, how they induce and regulate both adaptive immune responses and inflammation, especially in vivo, FcR humanized mice, and the multifaceted properties of FcRn. This book provides an essential overview of the role of phosphoinositide-3-phosphate kinase (PI3K) isoforms in modulating the function of immune system cells and their involvement in disease. PI3K is a family of kinases involved in basic cellular processes such as proliferation, migration and cell death. Recent work has highlighted the multiple roles of PI3K classes and subunits and their involvement in the immune response to the body's own and foreign antigens and diseases such as cancer and autoimmunity. Foot-and-mouth disease (FMD) has been recognized in printed records dating from the sixteenth century, and since the eradication of rinderpest (cattle plague) in the early part of the twentieth century it has been rec- nized as the most important and feared disease of cattle and other dom- tic livestock. The beginning of the twenty-first century brought the worst outbreak of FMD ever experienced in England, which had been completely free of the disease for 33 years. This tragic epidemic, which spread to Northern Ireland, Scotland, France and the Netherlands with severe e- nomic consequences, emphasized the need for further research into better methods for the detection and control of the disease. FMD is caused by a small RNA virus which is highly contagious and can survive in meat and other animal products for long periods at normal pH levels. The virus typically infects cloven-hoofed animals, including c- tle, goats, pigs and sheep, as well as a wide range of non-domesticated a- mals in regions of the world where FMD virus is endemic, such as the Af- can continent. There are seven recognized serotypes of FMD virus, with numerous subtypes, and as a consequence vaccine production and administration is complex and a major debate surrounds every disease outbreak regarding the relative merits of vaccination as opposed to the slaughter of all infected animals.

Lucille Cholerton is a specialist in gluten sensitivity and coeliac disease. She and her three grown children all suffer from gluten sensitivity. Scientific research on dengue has a long and rich history. The literature has been touched by famous names in medicine- Benjamin Rush, Walter Reed, and Albert Sabin, to name a very few- and has been fertile ground for medical historians . The advances made in those early investigations are all the more remarkable for the limited tools available at the time. The demonstration of a viral etiology for dengue fever, the recognition of mosquitoes as the vector for transmission to humans, and the existence of multiple viral variants (serotypes) with only partial cross-protection were all accomplished prior to the ability to culture and characterize the etiologic agent. Research on dengue in this period was typically driven by circumstances. Epidemics of dengue created public health crises, although these were relatively short-lived in any one location, as the population of susceptible individuals quickly shrank. Military considerations became as a major driving force for research. With the introduction of large numbers of non-immune individuals into endemic areas, dengue could cripple military readiness, taking more soldiers out of action than hostile fire. Dengue and dengue hemorrhagic fever, which assumed pandemic proportions during the latter half of the last century, have shown no indication of slowing their growth during this first decade of the twenty-first century. Challenges remain in understanding the basic mechanisms of viral replication and disease pathogenesis, in clinical management of patients, and in control of dengue viral transmission. Nevertheless, new tools and insights have led to major recent scientific advances. As the first candidate vaccines enter large-scale efficacy trials, there is reason to hope that we may soon "turn the corner" on this disease.
Natural killer T (NKT) cells are CD1d-restricted T cells that respond to lipid, not protein, antigens presented by CD1d, which is a non-classical MHC class I-like molecule. This relatively small but pivotal T cell population bridges the gap between innate and adaptive immunity by rapidly produce large amounts of many kinds of cytokines and by providing the T cell arm of the adaptive immune system a mechanism to detect lipid antigens. NKT cells also have both regulatory and effector functions and have been shown to play critical roles in the regulation of immune responses in many disease settings including cancer. Stimulating the protective functions of NKT cells while inhibiting the negative regulatory activity shows promise for the therapy of cancer and several approaches are in clinical trials. This book reviews what we have learned about NKT cells during last two decades through functional studies in the context of tumor immunology, including clinical trials of NKT cell targeted therapy. This knowledge should also be informative to understand the role of this underappreciated T cell population in many other fields outside of tumor immunology.

Vaccines against antigenically stable pathogens, or pathogens that only exist in a limited number of serotypes, have been very successful in the past and have drastically decreased the incidence and lethality of many diseases. However, when it comes to highly variable pathogens or viruses that exist in multiple serotypes, the traditional methods for vaccine development have reached their limits. This volume highlights the development of vaccines against such challenging pathogens. Novel approaches for immunogen design, including structure-guided vaccine development and vaccines targeting glycans, as well as adjuvants and animal models used for testing possible vaccine candidates are outlined and discussed in detail. Given its scope, the book will appeal to scientists in the fields of infectious diseases, microbiology and medicine.
This volume provides an overview on the influence of Extracellular Matrix (ECM) on tumor progression. It covers topics such as signaling induced by structural ECM proteins including collagen and fibronectin, the control of ECM deposition and the turnover in tumors. Also discussed are the migration of cells through basement membranes and the function of proteoglycans including lumican and veriscan in tumor progression. Chronic diseases are the leading cause of deaths worldwide and according to the World Economics Council and the Harvard School of Public Health, the cost of chronic diseases is expected to reach a staggering 48% of global gross domestic product by the year 2030. The urgency of the issue was demonstrated in 2011 when for only the second time in its existence, the U.N. General Assembly brought a health issue to the floor for consideration: chronic diseases. To date, most considerations of the issue have approached the topic from the vantage point that chronic diseases are a myriad of largely unconnected diseases and conditions arising in diverse tissues, organs and physiological systems. This book, "Immunotoxicity, Immune Dysfuction, and Chronic Disease," deviates from that prior model. It considers the interconnectivity of chronic diseases with both environmental insult of the immune system and subsequent immune dysfunction and inflammatory dysregulation as the underlying basis for many, if not most, chronic diseases. This change in the perception of environment-immune linkages to chronic disease is significant and has immediate implications both for the prevention of disease as well as for the development of more effective therapeutic approaches. Rather than considering environmental factors and types of reported immune alterations ("e.g., " depressed humoral immunity) as is common in books involving immunotoxicity, the present book approaches the environment-immune-disease triad from the standpoint of the disease. Each chapter emphasizes one or more specific immune dysfunction-based chronic disease(s) or condition(s) ("e.g., " asthma, atherosclerosis, multiple sclerosis, lupus) and describes: 1) the suggested environmental risk factors, 2) the underlying immune dysfunction(s) associated with the disease and 3) the overall health consequences of the disease. This book is an early entry for a new Toxicology book series for Springer titled: Molecular and Integrative Toxicology (MaIT). The series will feature detailed research information, but in the context of a more integrative or holistic framework. As part of this framework, the chapters will contain a section on Key Points as well as Recommendations where appropriate. The goal is to cover the most timely, state-of-the-art issues in toxicology as well as to ensure that the information is maximally accessible for research scientists, teachers, physicians and students.
