The purpose of Molecular Biology ofDiabetes, Parts I and II is to bring to these individuals the latest knowledge of diabetes-related research in a comprehensive, yet concise manner. To this end, we have assembled chapters, written by most of the world's experts in the field, that we believe compre hensively survey and synthesize a coherent understanding of the subject. Studies of the etiology of type I and type II diabetes are extremely exciting and essential, since we hope to one day prevent the disease using gene therapy. These aspects are covered in Molecular Biology of Diabetes: I. Autoimmunity and Genetics; Insulin Synthesis and Secretion. In type II diabetes, an abnormality in pancreatic secretion exists concomitantly with peripheral insulin resistance. This abnor mality of insulin secretion is believed to be related to a defect(s) in glucose sensing. Uncoupling of glucose sensing from insulin secre tion may be the crucial step in the pathogenesis of noninsulin-depen dent diabetes. In this volume, we have invited authors to describe their studies on all known factors affecting -cell function, including autoimmunity and genetics of diabetes, as well as molecular mecha nisms of insulin synthesis and secretion. In the last few years, the most rapidly advancing area of research in diabetes has been, in fact, related to insulin action."

1. 1 Experimental Diabetes in the Sand Rat 1. 1. 1 Zoology Experimental diabetes has been used in numerous animals, of which rodents have been predominant because of their ease of maintenance and handling (Herberg 1979), but some species of ape and monkey have also occasionally been used (Brunk 1971; Malaisse and Malaisse-Lagae 1970). Herberg et al. (1976) divided rodent species into two groups based on the similarity of their metabolic response to the induction of diabetes to the onset of diabetes mellitus in man. In one of these groups (mouse, Mongolian gerbil, Tuco tuca), the response of the metabolism to the induction of diabetes was similar to the changes seen in the onset of adult-type diabetes mellitus in man; while the other group, which included the sand rat, spiny mouse, Chinese hamster, and Djungarian hamster, responded to diabetes induction in a way suggestive of juvenile insulin-dependent diabetes. Such a subdivision is not without prob lems, however, particularly since difference occur not only between the various species of a group (Herberg et al. 1976), but, in our experience, also between animals of the same species (for instance the sand rat). On February 21 and 22, 1975, an International Workshop on the "Genetics of Diabetes Mellitus" was held in Goettingen, West-Germany. This workshop had been organized hy the Department of Medicine, University of Goettingen, and was generously sponsored by the Deutsche Forschungsgemeinschaft. Some forty geneticists and clinicians from Europe and N orth America, working in the field of diabetes both in man and laboratory animals, participated.
