The goal of Biological Aging: Methods and Protocols is to present some of the most promising and important tools that are currently used in biological aging research. These tools include established protocols such as aging cell culture as well as many more contemporary approaches such as nuclear transfer, microarray and proteomics technologies and the use of ribozymes in aging research. Collectively, these powerful tools combined with the many other techniques that are presented are rapidly advancing the exciting and expanding field of biological aging.

This volume is an outgrowth ofcontemporary research on development over the adult lifespan, which by now has burgeoned and developed both nationally and internationally. However, for us, the impetus to be involved in this area was spawned and nurtured by our initial association with the Society for Research in Adult Development (SRAD) with its origins some 15 years ago by Michael Commonsand his associates inCambridge, Massachusetts. Throughthegood will and support of this society, we also became, and are still, heavily involved with the Journal of Adult Development and the Kluwer-Plenum Monograph Series on Adult Development and Aging, ofwhich this volume is a companion. Many ofthe contributions in the volume are from SRAD members, who con sistently adhere to a focus on positive adult development. Their chapters have been complemented by pieces from other researchers, who have adopted more mainstream approaches to adult development and/oraging. Regardless ofthe par ticular approach and/or focus of the chapter, all the work reported herein sup ports the relatively recent idea that development is not restricted to children and adolescents but continues throughout the adult lifespan in ways that we never envisionedsome 20 years ago. Thus, the volume represents state-of-the-arttheory, research, and practice on adult development, which has the potential to occupy us all for some time to come."

Understanding the relationship between frailty and longevity becomes increasingly important as the world continues to age and life expectancy in most countries continues to increase. Recent studies have indicated that epigenetic processes may play a major role in both cellular and organismal aging. These epigenetic processes include not only DNA methylation and histone modifications, but also extend to many other epigenetic mediators such as the polycomb group proteins, chromosomal position effects, and noncoding RNA. The topics of this book range from fundamental changes in DNA methylation in aging to the most recent research on intervention into epigenetic modifications to modulate the aging process. The major topics of epigenetics and aging covered in this book are: 1) DNA methylation and histone modifications in aging; 2) Other epigenetic processes and aging; 3) Impact of epigenetics on aging; 4) Epigenetics of age-related diseases; 5) Epigenetic interventions and aging: and 6) Future directions in epigenetic aging research.

The most studied of epigenetic processes, DNA methylation, has been associated with cellular aging and aging of organisms for many years. It is now apparent that both global and gene-specific alterations occur not only in DNA methylation during aging, but also in several histone alterations. Many epigenetic alterations can have an impact on aging processes such as stem cell aging, control of telomerase, modifications of telomeres, and epigenetic drift can impact the aging process as evident in the recent studies of aging monozygotic twins.

Numerous age-related diseases are affected by epigenetic mechanisms. For example, recent studies have shown that DNA methylation is altered in Alzheimer s disease and autoimmunity. Other prevalent diseases that have been associated with age-related epigenetic changes include cancer and diabetes. Paternal age and epigenetic changes appear to have an effect on schizophrenia and epigenetic silencing has been associated with several of the progeroid syndromes of premature aging. The Last Years of Long Lives is a unique account of that period of old age which precedes death. Based on 400 complete individual histories and covering a twenty-year period, it looks at the experiences of people over eighty years old in three important areas: disability, family life and health care. Using the life-course approach to research, it reveals rich data about the contributions of formal and informal care and how life expectancy and experiences of disability interact with experiences of care.

The reader is invited to conceptualise these phenomena as processes in continuous time - processes that are sometimes long and complex, sometimes short and simple - and learns about the four types of disability career before death. In recent years, remarkable discoveries have been made concerning the underlying mechanisms of aging. In Life-Span Extension: Single-Cell Organisms to Man, the editors bring together a range of illuminating perspectives from researchers investigating the aging process in a variety of species. This novel work addresses the aging process in species ranging from yeast to man and, among other subjects, features detailed discussions of the naked mole-rat, an exceptionally long-lived rodent; the relationship between dietary factors/food restriction and aging; and an evolutionary view of the human aging process.

Single mutations that extend life span have been identified in yeast, worms, flies, and mice, whereas studies in humans have identified potentially important markers for successful aging. At the same time, it has been discovered that the genes and pathways identified in these studies involve a surprisingly small set of conserved functions, most of which have been the focus of aging research for some time. For example, the mTOR pathway, a regulator of translation and protein synthesis, has been identified as a common longevity pathway in yeast and Caenorhabditis elegans. The United Nations Madrid International Plan of Action on Ageing (MIPAA) offers a bold new agenda for handling the issue of ageing in the 21st-century. It focuses on three priority areas: older persons and development; advancing health and well-being into old age; and ensuring enabling and supportive environments.
