The occurrence of a negative energy balance (NEB) in dairy cows and its associated challenges for animal health and metabolism is one of hot topics in animal and veterinary sciences. Performance, metabolic and endocrine parameters were determined to study the effects of a NEB on adaptability in dairy cows. Commonly, dairy cows experience a NEB induced by lactation immediately post partum that may last up to 14 weeks of lactation. However, a NEB may also occur later in lactation during periods of insufficient feed quality and supply or during cases of illness. How do dairy cows adapt to energy deficient situations later in lactation? Are the mechanisms comparable to the metabolic adaptations post partum? Besides the lactation-induced NEB, responses during a deliberately induced energy deficiency by feed-restriction at around 100 days in milk were investigated. The different adaptive reactions of dairy cows to an energy deficiency at two stages in lactation indicate the different levels of metabolic priority of milk production during the course of lactation.

We can only control what we can measure. In the case of dairy farming, nitrogen utilisation requires a higher level of control to reduce environmental pollution and improve farm economics. Nitrogen metabolism in the ruminant animal is a complicated system and accurate evaluation of protein utilisation by dairy cows is limited by current methods. This dissertation assesses various methods of analysing ruminal crude protein degradation and introduces a new method of assessing protein value of forages i.e. the protein available to the ruminant for absorption at the duodenum. The following procedures are included: in situ incubation, Cornell Net Carbohydrate and Protein System, in vitro enzymatic degradation and the modified Hohenheim gas test. The third section of this dissertation applies these methods to a controlled experiment investigating the effect of wilting on the protein value, protein characteristics and amino acid profile of grass silage. Conclusively, the results of this study aid in estimating protein supply from forages to dairy cows.

252Cf Plasma desorption (PD), matrix-assisted laser desorption (MA-LDI) and electrospray ionization (ESI) time-of-flight (TOF) mass spectrometric techniques were invented few decades ago and have been used extensively to analyze large non-volatile bio-molecules by desorbing/ ionizing them into the gaseous phase. However, since the introduction of these techniques, the ion formation and matrix effects have been under constant debate. This monograph which is the outgrowth of my doctoral dissertation, describes ionization pathways and matrix-effects in PD-, LDI-TOF MS considering low- mass natural products below 1.0 kDa. The probable explosive matrix-assistance in PD-TOF-MS, via the explosive energy flow through explosive decomposition under the action of 252Cf fission fragment impact is also described. Furthermore, the potential of ESI Fourier transform ion cyclotron resonance MS coupled with MS/MS to characterize the individual components present in a mixture is demonstrated. This work also describes the construction details and preliminary results of the first MALDI-TOF mass spectrometer in Sri Lanka.

The present investigation was carried out to study the effect of the different processing treatments (blanching, sulfuring, freezing preservation and dehydration processes) on the chemical composition, enzyme activities and microbiological activities of the some vegetables(green peas, green beans and okra) during processing and storage. Results indicated that blanching treatment before freezing caused higher inhabitation of enzymes activities and load microbial in different vegetables depend on temperatures and time blanching. On the other hand, storage green vegetables frozen (peas, beans and okra) at -18 0C for 12 months caused slightly decreased after storage in chlorophylls, ascorbic acid and carotene content. Treatment of blanching before freezing led to great inhabitation effect of enzyme and reduce of load microbial in green vegetables compared with un-blanched during freezing. Also, blanching treatment protect the color, flavor and texture of samples especially samples blanched at 90 0C /5 minutes than other temperatures. The data indicated that both blanching and dehydration (sun or oven) for 12 months led to reduction of chemical composition and sensory evaluation.

This report represents an overview of the First National Nutrition Survey (FNNES2000). This survey was a voluntary cross-sectional population survey which provides information on food and nutrient intakes, daily diary on intake of food over one week, and nutrition-related clinical measures of Palestinians on a nationally representative sample of 3620 Palestinians living in selected households and aged 18 -64 years. Data were collected over the 12-month period October 1999 to October 2000. FPNNS2000 is the first and most comprehensive food and nutrition survey ever undertaken on the Palestinian population. The survey data were collected in the homes of respondents and included: a first and second 24 hour diet recalls; a daily diary on intake of foods over one week that included questions on food preparation habits; physical measurements including weight, height, hip to waist ratio; and blood pressure.

Food safety issue in the airline catering sector is an all encompassing issue that involves the food preparation from siting of the industry, to storage and handling by crew members. When food served on board is not produced and handled under strict hygienic conditions and following food safety rules, incidents of foodborne illnesses can result. Cases of foodborne disease outbreaks aboard commercial aircraft from 1947 to 2007 were analysed in this book with the focus on the vehicle of infection, causative agent and factors responsible for the outbreaks. The analysis of these outbreaks revealed that microbial infection is not directly related to the type of food served on board, but to weaknesses and failures in the system such as lack of compliance with basic food handling and general hygiene principles and also because HACCP principles were not in place when many of the outbreaks occurred, although a few outbreaks still occurred following the introduction of HACCP principles in this sector. The strict adherence to food safety as highlighted in this book will help prevent the reoccurrence of airline foodborne diseases outbreaks, otherwise an outbreak is inevitable.

Dairy production systems of 165 sample farms were examined to assess socio-economic status of producers, farm management practices and production constraints. Milk quality tests on milk of high grade Holstein type cows were also conducted to determine fat %, TS, SNF content of milk and prevalence of mastitis. The average family size was 6.4 persons per HH and 26.8 % of HHHs were females. The mean cattle herd size was 8.01 with the predominant breed being high-grade cattle. Among the three breeds Holestein type showed best reproductive and produvtive performance while the local ones performed least. Crossbred had respective performance values that lied between the two breeds. Parity and lactation stage significantly affected fat but not TS and SNF content of milk. Prevalence of mastitis differed significantly by parity and lactation stage It was thus, found out that there is a favorable environment for growth of the dairy sector that mainly comprised small producers and High-grade dairy type animals. The sector also contributes great roles in increasing household income of producers and the well being of city dwellers through supplying milk and milk products.
