Fatty acids play an important role in the barrier function of skin and represent a major source of proinflammatory mediators such as prostaglandins, leukotrienes and other lipids in inflammatory skin disorders. This book combines the two major functions of fatty acids in skin biology. In the first part the biosynthesis of fatty acids in skin with its role in barrier function as well as the role of dietary fatty acids on skin cell function and in the treatment of inflammatory skin diseases is presented. The second part deals with skin as a source of proinflammatory eicosanoids, especially with the keratinocyte as a major cellular source. Metabolism of eicosanoids in skin, its role in psoriasis and atopic dermatitis as well as pharmacological inhibition of eicosanoid biosynthesis is reviewed. The book finishes with a chapter describing the methods used for quantification of fatty acids and derivatives in skin inflammation. Anyone interested in skin physiology would benefit from the overviews about the two sites of fatty acids' function in skin integrity and in skin inflammation.
Epidermolysis bullosa (EB) is a heterogenous group of genodermatoses characterized by the formation of blisters and erosions on skin and mucous membrans from birth on. The cause are mutations in the genes of structural proteins of the junction between epidermis and dermis. This book deals with the treatment of this skin disease itself and its many extracutaneous complications. There is no previous book which has been focused on the therapy and it will be based on evidence-based data derived from the worlds largest cohort of inherited EB-patients, the American EB Registry. An important chapter will discuss gene therapy in hereditary EB which has been recently successfully performed within a localized skin site on a single EB patient as a proof-of-principle test. Given its unique collective contents, the monograph will provide the primary source for clinical informations of this oftentimes severe multiorgan disease."

effects of solar radiation. The physiological consequences of sun
exposure have been systematically studied starting at the end of
the nineteenth century and we now have accumulated knowledge about
how Caucasian and Asian skins reacts to solar radiation. The
chemical effects of solar ultraviolet radiation have been analyzed
with particular emphasis during the second half of the twentieth
century. Research on micro-organisms has allowed us to understand
the mechanisms of UV-induced mutagenesis and photosensitization.
Studies with laboratory rodents have opened the path to the
understanding of UV-induced immune-depression, carcinogenesis,
photo-damage and photo-aging. The results of these studies have
enabled other scientists to investigate the same phenomena in human
organs such as the skin and the eye. UV radiation damages hair, as
well. The present knowledge in these fields is summarized in some
of the chapters of this monograph.
Dermoscopy is a rapidly expanding imaging technique that has caught the fancy of clinicians and patients alike. Recent research has shown that BRAF inhibitors are effective for a short period of time, but there is little hope that this drugs as single agents will lead to durable benefit in a majority of patients. Among scientists and researchers who work in drug discovery, there is a lot of interest in the development of molecularly targeted cancer agents. Namely, the identification of a molecular target, the selection of molecules which effectively inhibit this target. What is starkly different about the development of this class of compounds, however, is that the mechanism of action of these agents are not as straightforward as was once previously assumed and the mechanisms of resistance that tumor cells employ to evade complete destruction are unlike any that have been described before. These discoveries in addition to utilization of modern molecular biology techniques have led to a series of hypotheses regarding which other types of molecules could be used in combination with BRAF-inhibitors in hopes of revolutionizing the potential of therapeutics in melanoma.
