Stability constants are fundamental to understanding the behavior of metal ions in aqueous solution. Such understanding is important in a wide variety of areas, such as metal ions in biology, biomedical applications, metal ions in the environment, extraction metallurgy, food chemistry, and metal ions in many industrial processes. In spite of this importance, it appears that many inorganic chemists have lost an appreciation for the importance of stability constants, and the thermodynamic aspects of complex formation, with attention focused over the last thirty years on newer areas, such as organometallic chemistry. This book is an attempt to show the richness of chemistry that can be revealed by stability constants, when measured as part of an overall strategy aimed at understanding the complexing properties of a particular ligand or metal ion. Thus, for example, there are numerous crystal structures of the Li+ ion with crown ethers. What do these indicate to us about the chemistry of Li+ with crown ethers? In fact, most of these crystal structures are in a sense misleading, in that the Li+ ion forms no complexes, or at best very weak complexes, with familiar crown ethers such as l2-crown-4, in any known solvent. Thus, without the stability constants, our understanding of the chemistry of a metal ion with any particular ligand must be regarded as incomplete. In this book we attempt to show how stability constants can reveal factors in ligand design which could not readily be deduced from any other physical technique.

Fluorescence spectroscopy continues its advance to more sophisticated methods and applications. As one looks over the previous decades, its appears that the first practical instruments for time-resolved measurements appeared in the 1970's. The instrumentation and analysis methods for time-resolved fluorescence advanced rapidly throughout the 1980's. Since 1990 we have witnessed a rapid migration of the principles of time-resolved fluorescence to cell biology and clinical appli- tions. Most recently, we have seen the introduction of multi-photon excitation, pump-probe and stimulated emission methods for studies of biological mac- molecules and for cellular imaging. These advanced topics are the subject of the present volume. Two-photon excitation was first predicted by Maria Goppert-Mayer in 1931, but was not experimentally observed until 1961. Observation of two-photon excitation required the introduction of lasers which provided adequate photon density for multi-photon absorption. Since the early observations of two-photon excitation in the 1960s, multi-photon spectroscopy has been limited to somewhat exotic applications of chemical physics, where it is used to study the electronic symmetry of small molecules. Placing one's self back in 1980, it would be hard to imagine the use of multi-photon excitation in biophysics or cellular imaging.

Mass spectrometry underwent dramatic changes during the decade of the 1980s. Fast atom bombardment (F AB) ionization, developed by Barber and coworkers, made it possible for all mass spectrometry laboratories to analyze polar, highly functionalized organic molecules, and in some cases ionic, inorganic, and organometallic compounds. The emphasis of much of this work was on molecular weight determination. Parallel with the development of ionization methods (molecular weight mass spectrometry) for polar biological molecules, the increased mass range of sector and quadrupole mass spectrometers and the development of new instruments for tandem mass spectrometry fostered a new era in structural mass spectrometry. It was during this same period that new instrument technologies, such as Fourier transform ion cyclotron resonance, radio frequency quadrupole ion trap, and new types of time-of-flight mass spectrometers, began to emerge as useful analytical instruments. In addi tion, laser methods useful for both sample ionization and activation became commonplace in almost every analytical mass spectrometry laboratory. In the last 5 years, there has been explosive growth in the area of biological mass spectrometry. Such ionization methods as electrospray and matrix-assisted laser desorption ionization (MALDI) have opened new frontiers for both molecular weight and structural mass spectrometry, with mass spectrometry being used for analysis at the picomole and even femto mole levels. In ideal cases, subfemtomole sample levels can be successfully analyzed. Sample-handling methods are now the limiting factor in analyz ing trace amounts of biological samples.

I knew nothing of the work of C. G. Vayenas on NEMCA until the early nineties. Then I learned from a paper of his idea (gas interface reactions could be catalyzed electrochemically), which seemed quite marvelous; but I did not understand how it worked. Consequently, I decided to correspond with Professor Vayenas in Patras, Greece, to reach a better understanding of this concept. I think that my early papers (1946, 1947, and 1957), on the relationship between the work function of metal surfaces and electron transfer reactions thereat to particles in solution, held me in good stead to be receptive to what Vayenas told me. As the electrode potential changes, so of course, does the work function at the interface, and gas metal reactions there involve adsorbed particles which have bonding to the surface. Whether electron transfer is complete in such a case, or whether the effect is on the desorption of radicals, the work function determines the strength of their bonding, and if one varies the work function by varying the electrode potential, one can vary the reaction rate at the interface. I got the idea. After that, it has been smooth sailing. Dr. Vayenas wrote a seminal article in Modern Aspects of Electrochemistry, Number 29, and brought the field into the public eye. It has since grown and its usefulness in chemical catalytic reactions has been demonstrated and verified worldwide.

Time-resolved fluorescence spectroscopy is widely used as a research tool in bioch- istry and biophysics. Berliner, eds. ) Modern techniques in proton NMR ofproteins (R. Krishna and L. J. Berliner, eds. ) Instrumental techniques of EPR (C. Bender and L. J. Berliner, eds. ) Thecurrent volume, Spin Labeling: The NextMillennium, presents an excellent collection of techniques and applications that evolved during the past decade since the last volume, volume 8 (1989). Someobvious omissions, such as multiquantum EPR and very high-frequency FT-ESR were unfortunately not possible for this volume. Perhaps they will appear in Spin Labeling: 2001. Lastly it is a pleasure to honor two scientists whose contributions were both pioneering and pivotal to the spin label technique: Professor Eduard G. Rozantsev (Moscow), whose synthetic feats in nitroxyl chemistry set the broad stage for a versatile catalog of labels; and Professor Harden M. McConnell, last year's Int- national ESR (EPR) Society Gold Medalist, who conceived and developed the spin label technique to address many biological problems (proteins, enzymes, m- branes, cells, immune response, etc. ).
